Research Description
We?re studying the gnenetic mechanisms underlying diabetic kidney disease (diabetic nephropathy or DN), the major cause of kidney failure in the U.S. Of people with diabetes mellitus, only a minority (~30%) get diabetic nephropathy. This is the result of an underlying genetic predispostion. Identification of the relevant genes in people has been slow because the mixed genetic background and diverse environmental factors. We are therefore characterizing the modifier genes causing DN in inbred mouse strains.
Roles for lipid mediators in the genesis of DN, especially cyclooxygenase derived prostaglandins, are another special area of interest.. All type II Diabetic patients are recommended to be on low dose aspirin to prevent cardiovascular problems. The importance of prostaglandins to systemic injury, is underscored by the fact that potent therapeutic drugs Motrin, Aspirin, and other Non-steroidal anti-inflammatory drugs (NSAIDs), all act by blocking the synthesis of prostaglandins via cyclooxygenases (COX1 or COX2). We are testing the hypothesis that changes in prostaglandin synthesis are integral to the development of DN, renal injury and regeneration of the injured kidney.
Finally, once synthesized prostaglandins activate several different families of receptors including nuclear receptors (PPARgamma) and G-protein coupled receptors. The roles of these different receptors are being studied in knockout mice with and without diabetes. Roles for prostaglandins in promoting renal regeneration, cell survival and stem cell differentiation are also being examined in the context of dehydration and diabetes mellitus. .
